The telomerase is an enzyme that is essential for cell replication and maintaining the integrity of chromosomes. It is responsible for the production and maintenance of telomeres, which are protective ends of chromosomes. Over time, telomeres naturally shorten with each cell division cycle, which can eventually lead to cell death or the development of age-related diseases.

CRITICAL ANALYSIS OF SCIENTIFIC LITERATURE ON TELOMERASE
Telomerase, an antiaging enzyme
During aging, gradual loss of telomeric DNA in the cell nucleus during DNA replication leads to senescence. Hence, accelerated shortening reveals many aging diseases and early death. A study tried to evaluate the connection between telomere length and mortality in 143 individuals aged more than 60. Those who had the shortest telomeres in the blood DNA had a lower chance of survival, which can be in part because of the cardiac mortality rate that is 3.18 times higher and the mortality rate that is 8.54 times higher in the case of infectious diseases. These results support the hypothesis according to which telomere shortening in the human person contributes to numerous aging diseases (1).
Zinc, an efficient agent for telomere quality

Pharmaceutical research indicates that astragalus extract component can improve telomerase activity and has antioxidant effects that neutralize free radicals that disturb cell functioning. It is also associated with anti-inflammatory and anticancer properties. Moreover, the extract is interesting because it is a lipid-lowering, hyperglycemic, and also hepatoprotective agent. Finally, astragalus extract has expectorant and diuretic properties. An astragalus dry-root extract, called TA-65, has been associated with a significant anti-aging effect on the immune system (4). Indeed, a recent 2021 study argues that cycloastragenol, a molecule present in astragalus, is currently the only natural telomerase activator (5).
Astragalus extract has proven its effects on humans
A study was led on 117 patients positive to cytomegalovirus, a virus of the herpes family. These patients were between 53 and 87 years old, and half of them received the astragalus extract, TA-65, which activates telomerase, and the other half received a placebo. This study summarizes the results on the length change of telomeres over a one-year period. It was shown that the patients taking the TA-65 intake significantly increased the length of their telomeres during the 12-month period, whereas those receiving the placebo significantly saw a reduction in telomere size. Therefore, the results suggest that TA-65 can statistically and clinically extend telomeres in a significant manner (6).

Demonstrated efficiency of astragalus extracts: cycloastragenol and astragaloside IV
It has recently been demonstrated that two small molecule telomerase activators, derived from astragalus, called astragaloside IV and cycloastragenol improve the lymphocyte T CD8 + proliferative response of people infected with HIV by regulating the activity surge of telomerase. T CD8 lymphocytes are cells that protect us from pathogens. They are part of our immune system (8).
Astragaloside IV, an astragalus extract has an effect against diabetic syndromes
Diabetic nephropathy is a complication that affects the kidney and that is a common symptom of diabetes. Astragaloside IV, an astragalus extract offers various pharmacological properties, especially a protective effect in diabetic people. In vivo and in vitro experiments have confirmed that astragaloside IV has oxidative anti-stress effects, hence against the harmful effects of free radicals on cells, but also anti-inflammatory ones. Astragaloside IV also plays an anti-epithelial-mesenchymal role. That means that it prevents the phenomenon by which kidney cells are forcefully transformed into fibrosis. Fibrosis contributes to kidney necrotizing because of the thickness of its tissue. Thanks to these properties, astragaloside IV improves renal lesions caused by high glycemia (9).
The cardio protective effect of astragalus extract
Cardiovascular diseases are the first cause of deaths in the world. Chinese traditional medicine has long proven to have a significant impact on the treatment of cardiovascular diseases. Astragalus is a traditional Chinese medicinal plant that is commonly used. Studies have shown that astragalus has anti-tumor properties, can regulate the metabolism, and reinforce immunity and the heart. Astragaloside IV is an active ingredient in astragalus, which plays a primordial role in cardiovascular diseases. It can protect against ischemic and hypoxic lesions, meaning artery narrowing. It can inhibit myocardial hypertrophy and myocardial fibrosis, hence the thickening and the narrowing of ventricular walls. It can improve myocardial contractility and diastolic disorders, meaning improving muscular function, attenuating vascular endothelial disorders, and fostering angiogenesis. This last phenomenon is associated with the creation of new vessels. Moreover, astragalus extract will also regulate glycemia and blood lipid levels and thus reduce the risk of cardiovascular diseases (10).
Telomerase has an effect on metabolic syndrome
The telomerase activator, a compound extracted from astragalus called cycloastragenol, is used in Chinese traditional medicine to extend life expectancy. A study led on 40 metabolic syndrome patients was made to evaluate the effects of the cycloastragenol molecule also called TA-65, on this at-risk population. Metabolic syndrome designates a set of consequences due to excessive weight fat. This study was led over the course of 12 weeks, during which half of the patients had to take 16 mg of a TA-65 supplement, and the others a placebo. The results were that those who had taken the TA-65 intake displayed higher HDL cholesterol, which is the “good cholesterol” that carries excessive cholesterol in the blood to the liver, where it will be destroyed. The study associates this positive change to an inflammation reduction. Furthermore, the body mass index, the waist size and the LDL/HDL ratio were reduced for those having taken TA-65 compared to those that took the placebo. Finally, there were fewer factors leading to tumor necrosis in those that received the TA-65 intake. The factors prompting tumor necrosis are proteins involved in the inflammation (11).
